Патофизиология данного процесса изучена не до конца, что осложняет разработку профилактических средств, раннюю диагностику и терапию начальных стадий заболевания. В связи с этим, моделирование ХБП в условиях отечественных научных центров является актуальной задачей российской фармакологии. Выполнение 5/6 нефрэктомии на крысах рекомендуется многими учеными как «золотой стандарт» для данной патологии. В настоящем исследовании была успешно сформирована ХБП у крыс Wistar модифицированным методом 5/6 нефрэктомии: лигирование сосудистого пучка целиком вместо отдельных ветвей почечной артерии. У двух групп животных (контроль и патология) проводили комплекс клиниколабораторных и патоморфологических исследований с целью определения критериев формирования ХБП. Повышение артериального давления и снижение клиренса креатинина позволили оценить прогрессирование заболевания у экспериментальных животных: к концу эксперимента данные изменения были частично обратимы, что характеризует выраженную адаптационную способность крыс. Для подтверждения формирования патологии выполняли гистологическую (гематоксилин и эозин, трихром по Массону) и иммуногистохимическую (CD68, Caspase-3) окраску срезов почек. В результате патоморфологического исследования были обнаружены характерные для ХБП изменения: участки апоптоза эпителия канальцев и образование фиброза со скоплениями CD68+ макрофагов. Таким образом были определены критерии формирования ХБП у крыс модифицированным методом 5/6 нефрэктомии.</p></abstract><trans-abstract xml:lang="en"><p>Chronic kidney disease (CKD) is a wide spread human illness defined as long-lasting abnormalities of kidney structure or function due to fibrosis and destruction of the normal architecture of the kidney. Pathophysiology of this process is not completely understood; thus, development of prophylactics, early detection and therapy of initial stage are complicated. Therefore, inducing of CKD in local research centers is a relevant objective for Russian pharmacology. 5/6 nephrectomy in rats is recommended by many scientists as “gold standard'” for this pathology. In this study CKD was successfully induced in Wistar rats using 5/6 nephrectomy, modified to meet the requirements of a large preclinical center. Two animal groups (control and pathology) underwent a complex of clinical diagnostic tests and pathological examination, so the CKD criteria could be specified. Elevated blood pressure and decreased glomerular filtration rate enabled assessment of the disease progress in experimental animals. At the end of the study these symptoms were partly reversible revealing prominent adaptability of rat kidney. In order to confirm pathology formation, we performed histological (hematoxylin-eosin, Masson's trichrome) and immonohistochemical (CD68, Caspase3) staining of kidney sections. During pathomorphological study we found alterations specific for CKD: apoptosis of tubule epithelium and fibrosis area crowded by CD68+ macrophages.
