В связи с тем, что в научной литературе нет данных о концентрации CYP450 в организме лабораторных крыс в зависимости от характера образцов тканей, основная цель нашего исследования включала изучение вариабельности уровня концентрации CYPOR в различном биоматериале (сыворотка крови, гомогенаты печени). Для исследования было сформировано несколько групп лабораторных крыс, весом 180-195 г., обоего пола (n=100). По окончанию карантинирования лабораторные крысы были эвтаназированы гуманным способом (ЛЭК при ФГБОУ ВО СПбГУВМ, протокол №2, от 20.04.2025 г.) с целью получения печёночного гомогената. Для этого пробы помещали в специальную среду суспендирования для гомогенизации тканей (pH=7,4; основа среды- трис-HCl буфер с добавлением 0,25М сахарозы и небольшого количества этилендиаминтетрауксусной кислоты и натриевой соли с целью связывания двухвалентных ионы металлов и глутатиона для препятствования переокислению липидных компонентов. Далее с помощью гомогенизатора (Stegler S10, Китай) получали 20 образцов гомогената (1 г ткани на 9 мл вышеуказанной среды), который анализировали методом иммуноферментного анализа. В результате исследования, полученные результаты в сыворотке крови были статистически не достоверны, и составили цифры интервалом от 0 до 470 нг/мл, и часто выходили за пределы чувствительности метода определения. В гомогенатах же печени была противоположная картина, где интервал был от 0,650 до 11,8 нг/мл, данные были репрезентативные, достоверные.</p></abstract><trans-abstract xml:lang="en"><p>A significant number of CYP450 enzymes of the mammalian body are localized in hepatocytes, which determines the high metabolic capacity of the liver in relation to most foreign substances. Due to the fact that there are no data in the scientific literature on the concentration of CYP450 in the body of laboratory rats depending on the nature of tissue samples, the main goal of our study included studying the variability of the CYPOR concentration level in various biomaterials (blood serum, liver homogenates). Several groups of laboratory rats weighing 180-195 g, of both sexes (n = 20) were formed for the study. At the end of the quarantine, the laboratory rats were humanely euthanized (LEC at the FSBEI HE SPbSUVM, protocol No. 2, dated 04/20/2025) in order to obtain liver homogenate. For this purpose, the samples were placed in a special suspension medium for tissue homogenization (pH=7.4; the basis of the medium is Tris-HCl buffer with the addition of 0.25 M sucrose and a small amount of ethylenediaminetetraacetic acid and sodium salt in order to bind divalent metal ions and glutathione to prevent peroxidation of lipid components. Then, using a homogenizer (Stegler S10, China), 20 homogenate samples were obtained (1 g of tissue per 9 ml of the above medium), which were analyzed by enzyme immunoassay. As a result of the study, the results obtained in the blood serum were statistically unreliable, and amounted to figures in the range from 0 to 470 ng / ml, and often went beyond the sensitivity of the determination method.
