Отбор образцов содержимого рубца осуществляли в зимне-весенний период в период забоя северных оленей на территории Мурманской области - в СХПК «Тундра» Ловозерского района. Всего было отобрано 12 образцов рубцовой жидкости. Образцы содержимого рубца, предназначенные для молекулярногенетических исследований, сразу после отбора были заморожены при температуре -20°С, а затем помещены на длительное хранение в морозильную камеру н Тотальную ДНК из образцов выделяли с помощью набора Genomic DNA Purification Kit («Fermentas, Inc. Литва»), в соответствии с рекомендациями производителя. Количество выделенной ДНК измеряли при помощи флуорометра Qubit® 2.0 (Life Technologies, США). Выделенную тотальную ДНК для дальнейшего анализа хранили при температуре -20°С. Оценку бактериального сообщества рубца проводили методом NGS-секвенирования на платформе для секвенирования следующего поколения MiSeq (Illumina, США) с применением праймеров для 16S рРНК. Секвенирование проводили при помощи реагентов для подготовки приготовления библиотек (Illumina, США). Обработку полученных ридов (прочтений ДНК), включающая перекрывание, фильтрацию по качеству (Q30), триммирование праймеров проводили с помощью биоинформатической платформы Illumina. Контроль качества и анализ данных проводился в соответствии с программой QlIME2 ver.2019.10 (https://docs.qiime2.org). NGS образцов рубцового содержимого северных оленей Мурманской области позволил описать закономерности бактериального биоразнообразия микробного сообщества рубца северных оленей и выявить ряд патогенных групп бактерий. Таким образом, использование метода NGS для анализа микробных сообществ рубца северного оленя представляется интересным для выявления различных патогенов и характеристики состояния пищеварительной системы и здоровья животного в целом.</p></abstract><trans-abstract xml:lang="en"><p>The method of next generation sequencing (NGS) allows deeply analyze the composition of the microorganisms of the rumen of ruminants. Sampling of the contents of the rumen was carried out in the winter-spring period during the slaughter of reindeer in the territory of the Murmansk region - in the agricultural complex “Tundra” of the Lovozero district. 12 samples of rumen content were taken. Samples of rumen contents intended for molecular genetic studies were frozen immediately after selection at -20 ° C and then placed for long-term storage in a freezer. Total DNA was extracted from the samples using the Genomic DNA Purification Kit (Fermentas Inc., Lithuania), in accordance with the manufacturer's recommendations. The amount of DNA extracted was measured using a Qubit® 2.0 fluorometer (Life Technologies, USA). The extracted total DNA was stored at -20 ° C for further analysis. Evaluation of the bacterial community of the rumen was carried out by NGS sequencing on the next generation sequencing platform MiSeq (Illumina, USA) using primers for 16S rRNA. Sequencing was carried out using reagents for library preparation (Illumina, USA). The processing of the reads (readings of DNA), including overlapping, quality filtration (Q30), and primer trimming, were performed using the Illumina bioinformatics platform. Quality control and data analysis was carried out in accordance with the QIIME2 ver.2019.10 program (https://docs.qiime2.org). NGS samples of rumen content of reindeer in the Murmansk region made it possible to describe the bacterial biodiversity patterns of the microbial community of the reindeer rumen and to reveal a number of pathogenic groups of bacteria.
